Human islets were cultured as monolayers on extracellular matrix; 4-to 9-day exposure to 33 mM glucose reduced islet insulin stores, insulin mRNA, insulin gene transcription, and the DNA-binding and mRNA levels of PDX-1, with no or minor changes in RIPE3b1 Act or IEF1. It is suggested that downregulation of PDX-1 plays a key role in glucose toxicity.
