This review presents in full the case for the distinction between T-lineage specification and commitment based on the timing of gene expression changes between thymocyte subsets of defined developmental potential. The function of transcription factors like GATA-1 and PU.1 that work as "central lineage regulators" is defined in terms of their effects on other transcription factors and on growth factor requirements, over and above their abilities to turn on lineage-specific target genes. This provides the context for a consideration of the regulatory pathways known to initiate B and T cell differentiation.

Previous studies, reviewed in [30], have shown that both EBF and E2A are essential for the earliest events in B-lineage specification. Here, O'Riordan and Grosschedl show that the dependence on both genes is so acute that a mixed heterozygote shows a developmental block almost as severe as a complete knockout of either one. The onset of expression of RAG-1, RAG-2, the pre-B cell receptor gene λ5, and Pax5 is severely inhibited. This paper also shows that EBF directly transactivates Pax5 (also see [32]).

This paper introduces a new gene-discovery approach to the search for T-lineage transcriptional regulators and presents a comprehensive analysis of the expression of genes of the Ets family during the course of T-cell differentiation. This analysis shows a combinatorial 'Ets code' distinguishing early B, NK, and T precursors at different stages. The results demonstrate both the uniquely dynamic, transitional regulatory phase associated with T-lineage specification and the highly selective repression of PU.1 expression coinciding with lineage commitment.

The lacZ detection is somewhat disappointing in sensitivity but the results are definitive enough to demonstrate that GATA-3 mutant precursors fail to populate the thymus with even the most primitive recognizable cell types.
