The p53-related peptide Glu-Pro-Pro-Leu-Ser-Gln-Glu-Ala-Phe-Ala-Asp-Leu-Trp-Lys-Lys (Promega) was used as substrate, and the kinase assay was performed as described (25), except that nuclear lysate (1 μl per 30 μl of reaction) was used instead of purified DNA-PK. To activate DNA-PK, we added sheared salmon sperm DNA at 100 ng per sample. The reaction was incubated for 30 min at 30°C, except for control samples, which were kept at 4°C, as indicated.
