11. 32P]ATP Either stereoisomer of DG64 was mixed with labeled DG71, DG67 (5′-GATCTGGCCTGTCTT-3′), and DG70 (5′-CTGCAGGGTTTTTGTTCCAGTCTGTAGCACTGT-3′) in 50 mM NaCl, heated to 90°C, and slowly cooled down to room temperature This mixture was then ligated with T4 DNA ligase (overnight at 16°C). The ligase was inactivated by incubation for 15 min at 65°C, and this oligonucleotide preparation was used directly in the cleavage assay In this substrate, the top strand is made up of oligonucleotides DG67 and DG71 and the bottom strand consists of DG70 and DG64 (reading from 5′ to 3′)
