The structures provide a rational for the specificity of the RalGEF and Raf-interacting Ras effector domain mutants.

    • Stimulation of gene induction and cell growth by the Ras effector Rlf
    • Together with the papers by Murai et al. [17] and Okazaki et al. [44], this study demonstrates the involvement of RalGEF-mediated signaling in transcriptional activation. An activated mutant of Rlf induces activation of the SRE , which can be inhibited by dominant negative Ral but not inhibitors of the Erk pathway. Finally, together with the paper from D'Adamo et al. [11], it describes that activation of RalGEF signaling alone is sufficient to induce serum-independent cell growth.
