    • New helper cells and matched early region 1-deleted adenovirus vectors prevent generation of replication-competent adenoviruses
    • To circumvent the formation of replication-competent adenoviruses (RCAs) during amplification in E1 complementing cell lines, a new complementing cell line PER.C6 containing Ad5 nucleotides 459-3510 was established. Propagation in this cell line of matched Ad vectors that lack these same Ad5 nucleotides leads to the undetectable generation of RCAs.

    • This paper reports that in vivo injection of a biologically inactive E1-deleted Ad leads to CTL induction. This observation demonstrated that a strong cellular response arose without de novo synthesis of either viral or transgene products, most probably because of the ability of viral particle proteins to enter the MHC-class I processing pathway. This study underlines the fact that even gutless vectors will not completely abrogate the host immune response and demonstrates the necessity of controling the immune system induction at the time of Ad injection.

    • Interaction of the adenovirus 14.7 kDa protein with FLICE inhibits Fas Ligand-induced apoptosis
    • This is the first demonstration that the E3-14.7 kDa protein binds to the effector caspase 8 common to both TNFR and Fas-apoptosis pathways. Previous studies involving the 14.7 kDa protein in the blocking of cPLA2 translocation should be reconsidered if the exact relationship between cPLA2 and caspases is to be unraveled.
