Science 1996, 274:373-376. This paper describes the use of a mutant adenovirus that has preferential replication advantage in p53-deficient human tumor cells in viral oncolysis. These results raise the possibility that mutant adenoviruses and other modified vectors may have potential in treating certain human tumors. This paper exemplifies the potential that adenovirus vectors will have in novel therapeutic uses.

J Virol 1997, 71:4626-4637. These results suggest that E1/E4-vectors have extended long-term persistence of viral DNA in the liver of lacZ transgenic mice. However, transient transgene expression was observed independent of the extent of the E4 deletion, suggesting that the choice of promoter may be critical to expression persistence.

J Virol 1997, 71:2408-2416. These results suggest that nonimmunological factors have a major influence on persistence of transgene expression. The choice of promoter is important for persistence of expression and some promoters are influenced by the context of the vector backbone.

Using mice genetically deficient in CD40 ligand, this study demonstrates that adenovirus transgene expression was stabilized and adenovirus-specific neutralizing antibodies failed to develop, permitting efficient repeat administration. Screening of a phage display peptide library for binding to the adenovirus fiber led to the discovery of fiber-binding peptides with limited homology to the MHC class I α2 domain.
