This paper shows that two regulatory proteins encoded by the genes CPH1 and EFG1 are involved in filamentous growth in both C. albicans and S. cerevisiae (STE12 is the homolog of CPH1 and PHD1 is the homolog of EFG1). The double mutants of these genes are defective in filamentous growth and the homozygous double mutant cph1 egf11 of C. albicans is nearly avirulent for mice. This study shows the importance in fungal pathogenicity of the conserved genes encoding regulatory proteins, rather than the species-specific genes.

In this study, the TUP1 gene, which encodes a transcriptional repressor, was isolated from C. albicans. It was shown that the tup1/tup1 mutant grows exclusively as filaments regardless of environmental conditions. The experimental model suggest that TUP1 represses genes responsible for initiating filamentous growth and the repression is lifted under some conditions, such as the presence of serum in the environment. Virulence data were not presented but the authors indicated that the tup1/tup1 mutant was less infectious in mice than the wild type. The animal model used may not have been appropriate for such strains.

The authors demonstrated not only that the conidial pigment of Aspergillus fimigatus is synthesized via the dihydroxynaphthalene-melanin pathway, but also that conidial pigment plays a role in virulence of the fungus. Further analysis identified the developmentally regulated gene alb1, which encodes a putative polyketide synthase; its disruption results in albino conidia with altered surface morphology. This is the first report of a virulence gene of A. fumigatus.
