J Clin Invest 1997, 99:2906-2914. This study showed for the first time a role for STAT5 in transcriptional regulation of ntcp. There was a time-dependent increase in nuclear translocation of liver STAT5 that correlated with serum prolactin levels. Binding of STAT5 to ntcp-GLEs was shown in electrophoretic mobility gel shift assay. A requirement for coexpression of PRLRL and STAT5 was shown by transient cotransfections in HepG2 cells. The short form of the PRL receptor failed to transactivate ntcp GLEs. These results support the conclusion that prolactin acting via PRLRL transcriptionally regulates ntcp by facilitating STAT5 binding to ntcp GLEs.

Demonstrates that during regeneration following hepatectomy, sinusoidal bile acid transport, ntcp mRNA, and ntcp mass are decreased without any effect on canalicular bile acid transport. These results indicate that the observed decrease in bile acid excretion following partial hepatectomy results from a decrease in bile acid uptake.

Hepatology 1997, 25:1176-1183. Shows that NTCP is downregulated in human patients with biliary atresia as it has been shown in rats with common bile duct ligation. More importantly, two patients with hypercholanemia showed no changes in NTCP expression or primary defect in the deduced amino acid sequence, raising the possibility that these patients may have a defect in a gene or genes other than NTCP.

Hepatology 1997, 25:1539-1542. A human homologue of rat cmoat, human cMOAT, was cloned and compared with the corresponding cDNA from fibroblasts of a patient with Dubin-Johnson syndrome. Results revealed a C- to-T mutation resulting in the conversion of the codon for arginine to a stop codon in the patient with Dubin-Johnson syndrome, thus providing an explanation for the transport defect in these patients.
