    • In spite of the minimal reactivity of cyclooctynes towards thiols, amines, and alcohols in vitro, it is interesting to note a recent report by Bertozzi and co-workers on the use of SPAAC for the labeling of biomolecules in live mice which resulted in the labeling of serum albumin with cyclooctynes (Ref. [76]). These authors have suggested that a nonspecific interaction of the hydrophobic cyclooctyne core with the protein might facilitate covalent bonding with cysteines or other nucleophilic residues. Further insight into this finding will help to elucidate the orthogonality of cyclooctynes. See also
