Attractor dynamics of network UP states in the neocortex
-
R. Cossart, D. Aronov, and R. Yuste Attractor dynamics of network UP states in the neocortex Nature 423 2003 283 288 The authors investigate the calcium dynamics of up to 1,400 cells in mice brain slices bathed in a Fura2-AM solution, revealing the occurrence of spontaneous synchronized high-frequency firing in coordinated networks of neurons. Cells involved in synchronized activity exhibit bimodal membrane potentials that simultaneously shift to suprathreshold UP (versus DOWN) states.

In vivo two-photon calcium imaging of neuronal networks
-
C. Stosiek, O. Garaschuk, K. Holthoff, and A. Konnerth In vivo two-photon calcium imaging of neuronal networks Proc Natl Acad Sci USA 100 2003 7319 7324 The authors use a patch pipette and a pressure ejection-based protocol to deliver locally a variety of AM esters of calcium sensitive dyes into mouse brain tissue in vivo. The ejection volume was ∼400 femtoliters, allowing the possibility of TPEF intracellular calcium response imaging of targeted neuronal networks with single cell resolution. The technique is suitable for mice of all ages.

Hyman Four-dimensional multiphoton imaging of brain entry, amyloid binding, and clearance of an amyloid-β ligand in transgenic mice Proc Natl Acad Sci USA 100 2003 12462 12467 TPEF microscopy reveals that Pittsburg compound B, which traverses the blood-brain barrier, is an excellent biomarker for the detection of amyloid-β plaques. This molecule can be readily tagged with a positron emission tomography contrast agent, paving the way for the early detection of Alzheimer's disease.

Water-soluble quantum dots for multiphoton fluorescence imaging in vivo

Webb Water-soluble quantum dots for multiphoton fluorescence imaging in vivo Science 300 2003 1434 1436 The authors show that water-soluble QDs are more efficient at producing TPEF by at least two orders of magnitude compared to the best standard water-soluble fluorescent dyes. These enable exceptionally deep imaging of capillaries in the skin tissue of living mice, and allow the possibility of multicolor emission imaging.

Brecht Targeted whole-cell recordings in the mammalian brain in-vivo Neuron 39 2003 911 918 The authors use TPEF from GFP labeled cortical interneurons to guide a pipette and enable targeted whole-cell patch recordings in vivo. In effect, GFP contrast is used here in vivo in a similar way as differential interference contrast (DIC) is used in standard electrophysiology experiments with slices. GFP labeling provides the added benefit that it facilitates identification of specific cellular populations.

3 regenerative amplifier
-
3 regenerative amplifier Opt Lett 28 2003 1022 1024 The authors further extend the principle of using pulsed excitation to produce TPEF. By amplifying laser pulse power at the expense of pulse repetition rate (keeping average power unchanged), they demonstrate a significant gain in depth penetration in brain tissue (see Figure 2). This strategy is appropriate when multiphoton induced photodamage is negligible and the only limit to depth penetration is insufficient laser power.

Webb In vivo multiphoton microscopy of deep brain tissue J Neurophysiol 91 2004 1908 1912 The authors demonstrate a panoply of TPEF neuroimaging possibilities with a needle-like GRIN lens, including in vivo microangiography with QDs, deep cortical imaging of yellow fluorescent protein marked neurons and axon bundles, and imaging of the hippocampal neuropil down to a depth of ∼1.5 mm. The insertion of the GRIN lens into the brain apparently leads to little tissue trauma.

Webb Uniform polarity microtubule assemblies imaged in native brain tissue by second harmonic generation microscopy Proc Natl Acad Sci USA 100 2003 7081 7086 SHG produces contrast in native unlabeled brain tissue, which the authors demonstrate to arise from microtubule assemblies in neuronal processes. Because SHG requires an asymmetric source, it is only generated when the individual tubulin strands in these assemblies exhibit the same polarity, allowing the identification of both structure and function of developing neuronal processes (see Figure 3).
