    • The orthogonal tilt reconstruction method: an approach to generating single-class volumes with no missing cone for ab initio reconstruction of asymmetric particles
    • The authors have developed a novel reconstruction scheme for single-particle EM that eliminates the missing cone problem of the more classical random conical tilt method. Such methodology could serve as a starting point to characterise the initial heterogeneity within a sample and to generate multiple references as templates for projection matching refinement.

    • The authors describe an elegant method for the estimation of the variance of a 3D reconstruction obtained using single-particle EM, which uses a bootstrap sampling technique. This and similar approaches will be crucial to establish the regions and sources of variability within the complex macromolecular structures implicated in DNA repair and other cellular macromolecular machinery. This method will also help to define areas within a large complex implicated in movements related either to flexibility or to alternation between several functional conformations.

    • Three-dimensional structure and regulation of the DNA-dependent protein kinase catalytic subunit (DNA-PKcs)
    • This work provides a cryo-EM reconstruction of DNA-PKcs at ∼15 Å resolution, sufficient to model several regions of this kinase implicated in NHEJ. DNA-dependent activation of the kinase at the site of the lesion is proposed to involve a structural rearrangement between the N-terminal HEAT-repeat-containing domain and the C-terminal kinase domain after DNA binding. This reorganisation of the molecule acts in combination with the repositioning of the FAT and FATC domains. They are directly linked to the catalytic domain and are proposed to regulate the conformation and activation of the kinase.
