    • A functionally orthogonal ligand-receptor pair created by targeting the allosteric mechanism of the thyroid hormone receptor
    • By mimicking known mutations found in resistance to hormone diseases, a crucial amino acid residue within TRβ is mutated that fosters an allosteric interaction necessary for coactivator recruitment. The resulting loss of function is subsequently rescued with a designed mutant-selective analog displaying high potency and orthogonal specificity, as displayed with genetic assays across multiple response elements.

    • The authors used directed evolution to engineer proteins of distant ligand recognition function by step-wise selection for recognition of ligands that gradually increase in target-like properties. By coupling evolved protein activity (in the form of transgene activation of a selection marker) with positive ligand-receptor interactions, an estrogen receptor was evolved towards responsiveness with testosterone; this receptor was further evolved towards progesterone activity, which, in turn, was additionally evolved towards cortisone responsiveness.
