Blood (2005) 106(9):2936-2943. Costimulation is important for T cell activation. It has therefore been proposed that the lack of CD80/86 is the direct cause of tolerance induced by immature dendritic cells. However, there has been little firm experimental evidence available to directly support this hypothesis. To help clarify this, this paper brings about the novel strategy of binding and destroying the CD80/86 in dendritic cells before its expression on the cell surface. The work offers novel support for the hypothesis that the absence of CD80/86 expression can render dendritic cells tolerogenic. From a therapeutic standpoint, the work also suggests a novel strategy that might render human monocyte-derived dendritic cells tolerogenic in vivo.

This is an important paper that highlights the deficiencies of viral vectors. It shows that viral vectors can activate immature dendritic cells through the activation of NF-κB, phosphatidylinositol 3-kinase (PI3K) and double-stranded (ds)RNA-triggered protein kinase (PKR) pathways. On the other hand, exposure of mature dendritic cells to viral vectors can also upregulate IDO activity and modulate the immunostimulatory capacity of mature dendritic cells.

(2006) 26(3):462-467. Using endothelial cells as a model, this work demonstrates that viral vectors are more efficient than non-viral vectors at transferring genes. However, the viral vectors can activate NF-κB, PI3K and dsRNA-triggered/PKR pathways. In addition, the paper also shows that blockade of the NF-κB, PI3K or PKR signalling pathways all operated to inhibit partially virally induced activation, and inhibition of both PKR and PI3K pathways totally blocked endothelial cell activation.
