    • Stimulation of cell proliferation by endosomal epidermal growth factor receptor as revealed through two distinct phases of signaling
    • This study and [13,14] provide functional insights into EGF and PDGF signalling from endosomes. Inactive receptors bound to ligands are internalised in the presence of specific inhibitors and triggered to signal from endocytic compartments after the removal of inhibitor. Such intracellular activation is sufficient to stimulate cell survival and proliferation.

    • Regulation of angiotensin II type 1A receptor intracellular retention, degradation and recycling by Rab5, Rab7 and Rab11 GTPases
    • This paper shows how endocytic Rab proteins co-operate in modulating the intracellular routing of angiotensin II type 1A receptor, a member of the GPCR family. Rab5 and β-arrestin binding to the AT1AR carboxyl-terminal tail mediates the retention of the receptor in early endosomes, while Rab7 and Rab11 regulate its targeting to late or recycling endosomes, respectively.

    • NGF signaling in sensory neurons: Evidence that early endosomes carry NGF retrograde signals
    • This is an impressive biochemical characterisation of endosomes carrying NGF signals, after their isolation from an intact nerve preparation. After placing a piece of rat sciatic nerve in a chamber, vesicles travelling in an antero- or retrograde direction were collected from both ends. The retrograde NGF- and TrkA-containing vesicles were shown to possess the markers of early endosomes and a set of active signalling molecules, lending strong support to the signalling endosome hypothesis.

    • This paper uses compartmentalised cultures of sympathetic neurons to demonstrate the requirement for the NGF internalisation and kinase activity of its receptor, TrkA, in both distal axons and cell bodies to support neuronal survival. Interestingly, TrkA activity, via recruitment of local adaptor proteins, may have different functions in distinct intracellular compartments.

    • Distinct endocytic pathways regulate TGF-β receptor signalling and turnover
    • This study proposes the concept that targeting of signalling receptors to different endocytic pathways can influence signalling output. It is reported that caveolae-mediated internalisation of TGF-β receptors into caveosomes does not lead to signalling, whereas clathrin-mediated internalisation into early endosomes allows downstream signalling.

    • APPL proteins link Rab5 to nuclear signal transduction via an endosomal compartment
    • This study identifies APPL1 and APPL2, two novel Rab5 effectors, which reside on a subpopulation of endosomes and undergo regulated nuclear translocation in response to extracellular stimuli. These findings indicate that endosomes are required to transduce certain proliferation signals and that specialised endocytic compartments devoted to signalling may exist.
