This paper reports the tissue expression patterns and intracellular localization of the two major variants of ORP1, and delineates determinants of the late endosomal targeting of the ORP1 long (L) variant. Especially ORP1L, but also certain other ORPs are shown to be upregulated during monocyte differentiation into macrophages, and ORP1L is found to enhance the action of liver X receptors. The data suggest a distinct function of ORPs in macrophage sterol metabolism.

The ceramide transport and sphingomyelin synthesis defect in a mammalian cell line is reported to be caused by a mutation in the CERT/GPBP gene, which encodes a protein highly similar to ORPs in domain structure. This protein has a Golgi-targeting pleckstrin homology domain closely related to that of OSBP and an endoplasmic reticulum-targeting FFAT motif, but instead of a carboxy-terminal ORD it has a START domain that is shown to bind ceramide. The CERT-mediated ceramide transport is suggested to occur at endoplasmic reticulum-Golgi membrane contact sites.

A novel short motif called FFAT is identified in a number of ORPs and other lipid binding proteins that is shown to target proteins to the endoplasmic reticulum. This occurs via a highly conserved direct interaction with homologues of VAMP-associated proteins, resident trans-membrane proteins of the endoplasmic reticulum. The authors suggest that the function of the FFAT motif may be to facilitate nonvesicular transport of newly synthesized lipids from the endoplasmic reticulum to other organelles.
