Systematic optimization of a lead-structure identities for a selective short peptide agonist for the human orphan receptor BRS-3
-
Weber D, Berger C, Heinrich T, et al.: Systematic optimization of a lead-structure identities for a selective short peptide agonist for the human orphan receptor BRS-3. J Pept Sci 2002, 8:461-475. This study investigates further the structural requirements of a previously identified synthetic peptide selectively binding to the human BRS-3. A tripeptide with selective agonist properties on the human BRS-3 is identified.

Elevated serum progastrin-releasing peptide (31-98) in metastatic and androgen-independent prostate cancer patients
-
Yashi M, Muraishi O, Kobayashi Y1 et al.: Elevated serum progastrin-releasing peptide (31-98) in metastatic and androgen-independent prostate cancer patients. Prostate 2002, 51:84-97. This clinical study showed that the serum proGRP levels were positively correlated with the prostate-specific antigen values in patients with untreated prostate cancer. ProGRP values increased transiently when the cancer progressed to an androgen-independent state. It was suggested that GRP may be a growth factor potentially upregulated by androgen.

Human gastrin-releasing peptide receptor mediates sustained CREB phosphorylation and transactivation in HuTu 80 duodenal cancer cells
-
Qu X, Xiao D, Weber H: Human gastrin-releasing peptide receptor mediates sustained CREB phosphorylation and transactivation in HuTu 80 duodenal cancer cells. FEBS Lett 2002, 527:109. Gastrin-releasing peptide receptor activation stimulated sustained CREB phosphorylation and transactivation in duodenal cancer cells through a PKC-dependent and partially p38 MAPK-dependent pathway. This novel signaling mechanism may be of importance for regulation of CREB-dependent gene expression in human cancers expressing functional GRP-R.

The metalloprotease Kuzbanian (ADAM10) mediates the transactivation of EGF receptor by G protein-coupled receptors
-
Van Y, Shirakabe K, Werb Z: The metalloprotease Kuzbanian (ADAM10) mediates the transactivation of EGF receptor by G protein-coupled receptors. J Cell Biol 2002, 158:221-226. This elegant article for the first time elucidates how metalloprotease-mediated shedding regulates signaling pathways and provides novel insights on EGFR transactivation mediated by G-protein-coupled receptors. It determines that ADAM 10(KUZ) is involved in GPCR-mediated Ras and Erk activation via EGFR transactivation.

Selective targeting and inducible destruction of human cancer cells by retroviruses with envelope proteins bearing short peptide ligands
-
Gollan TJ, Green MR: Selective targeting and inducible destruction of human cancer cells by retroviruses with envelope proteins bearing short peptide ligands. J Virol 2002, 76:3564-3569. This report provides a novel strategy to target selectively human cancer cells expressing functional GRP-R by modifying a retroviral envelope protein with short GRP peptides. Pseudotype vims transduces successfully target cells with cognate GRP-R and causes cell death via delivery of thymidine kinase activity in the presence of ganciclovir.
