Spectrum of PTCH1 mutations in French patients with Gorlin syndrome
-
Boutet N, Bignon YJ, Drouin-Garraud V, et al. Spectrum of PTCH1 mutations in French patients with Gorlin syndrome. J Invest Dermatol 2003; 121:478-481. These authors propose that mutation c1208-1209delAT is the first truly recurrent mutation identified in the PTCH gene. Patients bearing this mutation have a different clinical outcome, which pinpoints the possible involvement of modifier genes.

The magnitude of hedgehog signaling activity defines skin tumor phenotype
-
Grachtchouk V, Grachtchouk M, Lowe L, et al. The magnitude of hedgehog signaling activity defines skin tumor phenotype. EMBO J 2003; 22:2741-2751. These authors suggest that the levels of HH pathway activation and G1 cyclins are determinants of tumor phenotype, and implicate deregulated hedgehog signaling in the genesis of human basaloid follicular hamartomas. Expression of an activated SMO mutant in keratinocytes appears to be insufficient for the development and/or maintenance of full-blown BCCs.

Loss of protein kinase Cα expression may enhance the tumorigenic potential of Gli1 in basal cell carcinoma
-
Neill GW, Ghali LR, Green JL, et al. Loss of protein kinase Cα expression may enhance the tumorigenic potential of Gli1 in basal cell carcinoma. Cancer Res 2003; 63:4692-4697. Two protein kinase C (PKC) isoforms are expressed in BCC and hair follicles. PKCα negatively regulates Gli1 activity. This is important because PKCα is downregulated in BCC, whereas Gli1 is expressed in BCC.

Identification of a small molecule inhibitor of the hedgehog signaling pathway: Effects on basal cell carcinoma-like lesions
-
Williams JA, Guicherit OM, Zaharian BI, et al. Identification of a small molecule inhibitor of the hedgehog signaling pathway: Effects on basal cell carcinoma-like lesions. Proc Natl Acad Sci USA 2003; 100:4616-4621. A patent application, describing the use of cyclopamine to selectively block the Hedgehog pathway for therapeutic purposes, was filed by the Johns Hopkins University School of Medicine and subsequently licensed to CURIS (2003). CURIS has a number of interests in developing drugs that either promote or inhibit signaling activity in the Hedgehog pathway.

Omithine decarboxylase is a target for chemoprevention of basal and squamous cell carcinomas in Ptch1-t-/- mice
-
Tang X, Kim AL, Feith DJ, et al. Omithine decarboxylase is a target for chemoprevention of basal and squamous cell carcinomas in Ptch1-t-/- mice. J Clin Invest 2004; 113:867-875. α-difluoromethylomithine (DFMO) appears to be a useful inhibitor of ODC and, at least when used topically, appears safe. Mouse and human consumption of DFMO in cancer-chemoprevention studies has revealed no toxic effects, other than ototoxicity at high doses, which was reversible after discontinuation.
