    • The role of apoptosis in creating and maintaining luminal space within normal and oncogene-expressing mammary acini
    • By studying how lumen formation is maintained in a 3D tissue structure, the authors were able to implicate a role for autophagy in apoptosis regulation. The experiments emphasize the complex nature of 3D tissues and show how resistant tissues can be to malignant transformation. They demonstrate that lumen filling recapitulates an early stage of malignant transformation that requires both enhanced cell survival and increased proliferation. Moreover, although overexpression of an activated ErbB2 could induce a 'premalignant' phenotype, complete malignant transformation and invasion could not be induced.

    • Akt activation disrupts mammary acinar architecture and enhances proliferation in an mTOR-dependent manner
    • This is an interesting study examining the role of AKT in epithelial survival and morphogenesis using a reconstituted ECM assay and immortalized mammary epithelial cells. The data indicate that although AKT does have a modest and significant effect on cell survival it primarily induces cell growth and compromises morphogenesis.

    • Using 3D ECM assays and a unique tumor progression model that could be phenotypically reverted to recapitulate a 'polarized-differentiated' tissue structure, a role for tissue polarity in generating apoptosis resistance in cells was revealed. Tissue polarity and apoptosis resistance were shown to depend upon α6β4 integrin ligation and activation of NFκB, revealing a mechanism whereby the tissue microenvironment can modulate drug responsiveness of primary and metastatic tumors. These data explain why tumors in vivo that express α6β4 integrins and basement membrane proteins often have such an unfavorable prognosis.
