    • Regulation of anti-phosphatidylserine antibodies
    • Anti-DNA/phosphatidylserine B cells in transgenic mice appear regulated by receptor editing, with evidence for association of the transgenic H chain with a limited repertoire of endogenous L chains. Importantly, this study highlights the crossreactive nature of anti-DNA Abs and the difficulty in identifying the in vivo tolerogen. Moreover, it supports the hypothesis that anti-dsDNA B cells may be tolerized by encounters with Ags on apoptotic cells.

    • Delayed apoptotic cell clearance and lupus-like autoimmunity in mice lacking the c-mer membrane tyrosine kinase
    • C-mer is a tyrosine kinase involved in the recognition and clearance of apoptotic debris. Mice lacking the intracellular domain of c-mer have increased titers of anti-dsDNA auto-Abs, providing more evidence for the link between defects in apoptosis/apoptotic clearance and anti-dsDNA Ab production. Immunization with apoptotic cells does not accelerate anti-dsDNA auto-Ab production, indicating that endogenous apoptotic cells generated in the mice themselves are sufficient for auto-Ab production.

    • The efficiency of B cell receptor (BCR) editing is dependent on BCR light chain rearrangement status
    • This paper studies six different transgenic mice - each expressing a germline or somatically mutated anti-dsDNA H chain, combined with one of three different L chains (Vκ1-Jκ1, Vκ4-Jκ4, Vκ8-Jκ5). B cells in these mice have varying outcomes of receptor editing or anergy, with the efficiency of L-chain editing apparently related to the physical position of the utilized Vκ gene segment.

    • H + L chain Ig transgenic mice produce anti-Sm B cells with a lower affinity than anti-Sm B cells from H chain only transgenic mice. The B cells are phenotypically mature and of the B-2 subtype (in contrast to anti-Sm B cells in the H chain only model [51]), but appear functionally inactive as measured by decreased responsiveness to LPS in vitro. These data highlight the diverse outcomes revealed using distinct anti-Sm transgenic models.

    • Evolution of autoantibody responses via somatic hypermutation outside of germinal centers
    • AM14 Tg rheumatoid factor (RF) B cells in lpr/lpr mice differentiate into Ab-forming cells (AFCs). Although there is no evidence for the RF B cells localizing in germinal centers (GCs), they are found proliferating at the bridging channels to the red pulp. Remarkably, microdissection and analysis of the AFC clusters reveals extensive somatic hypermutation in the RF Ig genes at this extra-GC site. In this way, autoreactive B cells may escape the normal processes of negative selection that occur in a structured germinal center.

    • Immune complexes (ICs) composed of nucleosomes and IgG Abs are stimulatory for RF B cells, as opposed to ICs that lack Toll-like receptor (TLR) ligands. It is further shown that this response is dependent on MyD88 and probably acts through the TLR9 pathway. This is a novel demonstration of how mammalian DNA can be rendered stimulatory via IC internalization and delivery to TLR9. It is hypothesized that the dual engagement of BCRs and TLRs may be a mechanism for SLE-associated autoreactive B-cell activation.

    • Resistance to CpG DNA-induced autoimmunity through tolerogenic B cell antigen receptor ERK signaling
    • In vitro culture with CpG can induce Ab-forming cell (AFC) formation in both naïve and anergic cells from the hen egg lysozyme (HEL) B-cell tolerance model. However, when chronically stimulated with HEL Ag, anti-HEL B cells are prevented from forming AFCs in the presence of CpG. This may reveal a mechanism by which anergic cells undergoing chronic Ag stimulation are prevented from differentiating into AFCs in response to Toll-like receptor signaling.

    • Purified Ag-stimulated B cells are unable to stimulate Th1 cells to produce IFN-γ or induce a Th1-like phenotype in naïve T cells. However, if the Ag is conjugated with CpG, the B cells express increased levels of CD86, CD40, and MHC class II, secrete IL-12, and can induce T-cell differentiation into a Th1 phenotype. Thus, Ags containing TLR9 ligands may not only activate autoreactive B cells but also indirectly influence the direction of the associated T-cell response.
