Binder CJ, Chang MK, Shaw PX, et al. Innate and acquired immunity in atherogenesis. Nat Med 2002; 8:1218-1226. These reviews provide a comprehensive overview of the role of immune mechanisms in atherogenesis. In addition to the evidence that these may impact lesion formation, the authors suggest novel therapeutic approaches to modulate atherogenesis.

Minimally modified LDL binds to CD14, induces macrophage spreading via TLR4/MD-2, and inhibits phagocytosis of apoptotic cells
-
Miller Y, Viriyakosol S, Binder CJ, et al. Minimally modified LDL binds to CD14, induces macrophage spreading via TLR4/MD-2, and inhibits phagocytosis of apoptotic cells. J Biol Chem 2003; 278:1561-1568. This paper shows that mmLDL affects the cytoskeletal organization of macrophages mediated by CD14 and TLR4. The mmLDL-induced spreading of macrophages inhibits their ability to phagocytose apoptotic cells but enhances the uptake of oxidized LDL.

Protective role of phospholipid oxidation products in endotoxin-induced tissue damage
-
Bochkov VN, Kadl A, Huber J, et al. Protective role of phospholipid oxidation products in endotoxin-induced tissue damage. Nature 2002; 419:77-81. Oxidized PC-containing lipids are known for their pro-inflammatory effects. This paper, however, suggests that these oxidized lipids protect from adverse lipopolysaccharide-induced effects, both in vitro and in vivo. The authors explain these findings by the competition between oxidized lipids and lipopolysaccharide for CD14 binding.

Specific phospholipid oxidation products inhibit ligand activation of Toll-like receptors 4 and 2
-
Walton KA, Cole AL, Yen M, et al. Specific phospholipid oxidation products inhibit ligand activation of Toll-like receptors 4 and 2. Arterioscler Thromb Vasc Biol 2003; 23:1197-1203. This paper reports that OxPAPC inhibits LPS-induced TLR4-dependent secretion of IL-8 and MCP-1 in endothelial cells. It also describes a novel mechanism that involves alterations to lipid raft/caveolar processing, by which specific phospholipid oxidation products inhibit activation by TLR4 and TLR2 ligands.

A CD36-initiated signaling cascade mediates inflammatory effects of beta-amyloid
-
Moore KJ, El Khoury J, Medeiros LA, et al. A CD36-initiated signaling cascade mediates inflammatory effects of beta-amyloid. J Biol Chem 2002: 277:47373-47379. This paper shows that CD36 is not only a scavenger receptor but also initiates a signaling cascade mediated by beta-amyloid binding. It is not yet known whether OxLDL binding to CD36 also initiates such signaling.
