    • DNA shuffling of subgenomic sequences of subtilisin
    • Molecular breeding of a pool of 26 subtilisin sequences (56.4-99.5% DNA sequence identity) was analyzed for improved thermostability, function in organic solvent, and pH-dependence of the enzymatic activity. The best performing hybrids contained fragments from up to 25 of the 26 parents. The data also indicated that parents with the highest sequence homology to the chimeras were not necessarily the best starting point for directed evolution.

    • A combinatorial approach to hybrid enzymes independent of DNA homology
    • Functional hybrides from an ITCHY library of glycinamide ribonucleotide formyltransferases from E. coli and H. sapiens (50% DNA sequence identity) were compared to chimeras generated by DNA shuffling. Seventeen unique crossovers were identified by ITCHY, being distributed over the truncated region with no preference for regions of high homology. In comparison, only five unique crossovers were found by DNA shuffling, all concentrated in regions with high sequence homology.
