This cogent review discusses programmed cell death (apoptosis) as a major mechanism for the regulation of cell numbers and as a defense mechanism to remove unwanted and potentially dangerous cells. Two main mechanisms of action have been proposed to connect Bcl-2 to caspases. In the first one, antiapoptotic Bcl-2 maintain cell survival by dragging caspases to intracellular membranes and by preventing their activation. In the second, Bcl-2 acts by regulating the release from mitochondria of caspases activators: cytochrome c and/or AIF (apoptosis-inducing factor). This crucial position of mitochondria in programmed cell death control is reinforced by the observation that mitochondria contribute to apoptosis signaling via the production of reactive oxygen species.

The rate-limiting enzyme in dopamine synthesis, tyrosine hydroxylase (TH), is a selective target for nitration following exposure of PC12 cells to either peroxynitrite or 1-methyl-4-phenylpyridinium ion. Nitration of TH also occurs in mouse striatum after 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) administration. Nitration of tyrosine residues in TH results in loss of enzymatic activity. The resemblance of the MPTP model with Parkinson's disease (PD) suggests that a similar phenomenon may occur in PD, influencing the severity of Parkisonian symptoms.

Subsequent formation of nitrotyrosine was detected. The presence of superoxide dismutase had no effect on nitration, indicating that peroxynitrite was not the nitrating agent. Peptide sequencing indicated that a single modified residue was Tyr385, the source of the putative catalytically active tyrosyl radical.

The ortho and meta isomers, although of significantly different in vitro SOD activities, have close in vivo SOD efficacy due to their similarly weak interactions with DNA.
