The structure of NO synthase oxygenase domain and inhibitor complexes
-
Crane BR, Arvai AS, Gachhui R, Wu C, Ghosh DK, Getzoff ED, Stuehr DJ, Tainer JA: The structure of NO synthase oxygenase domain and inhibitor complexes. Science 1997, 278:425-431. The structural characterization of the monomeric heme domain in inducible nitric oxide synthase (iNOS) provides critical information about the active site for arginine binding and oxidation. The large substrate-binding pocket in this protein accommodates two imidazoles, one of which is bound directly to the iron. The next challenge in the structural characterization of this protein is a high resolution X-ray diffraction structure of the dimer of the heme domain.

In vivo aspects of (NO) chemistry: Does peroxynitrite (-OONO) play a major role in cytotoxicity?
-
Fukuto JM, Ignarro LJ: In vivo aspects of (NO) chemistry: does peroxynitrite (-OONO) play a major role in cytotoxicity? Accounts Chem Res 1997, 30:149-152. A critical appraisal of peroxynitrite's physiological role with an attempt to integrate the biological results with known chemistry. Perhaps an equally important question posed by this article, and one that deserves equal scrutiny, is: Does the nitroxyl anion have anything to do with the aqueous solution chemistry of NO?
