The most recent hemocyanin X-ray structure is presented, the first from a mollusc. Compared with the previous arthropodal structure [21], similar dicopper-binding sites and side-on dioxygen coordination are observed. One difference is that molluscs have an unusual thioether bridge between a histidine ligand and a nearby cysteine residue.

J Biol Chem 1998, 273:7957-7966. Methane monooxygenase (MMO) is found in methanotrophs, where it catalyzes the conversion of methane to methanol using dioxygen as co-substrate. A cytoplasmic soluble MMO possesses a nonheme diiron protein (see [47,57,58]), but a membrane-bound (particulate) p-MMO is copper(I) dependent. The isolation, purification and characterization of one p-MMO is described in this paper, showing that there are three kinetically distinct forms of the enzyme. Prior studies have provided insights into the metal environments, and the presence of sets of copper ion clusters has been suggested.

    • V = O). For a hydroxlyation reaction, this reactive intermediate abstracts an H-atom from substrate and rebounds (see also [65,67•]) to give hydroxylated product. This paper suggests (and offers a selection of other references) an alternative, since the authors show that a side-on bound peroxo-Fe(III) complex is a powerful nucleophile. They demonstrate two different selectivities during reaction with androgen-like substrates. When these react in the aldehydic form, the reaction product is an epoxide (i.e. P-450-like reactivity), whereas when the substrate form is stabilized in the enolic form, an oxidative aromatization occurs (i.e. aromatase-like reactivity, the aromatases being a subclass in the P-450 family, catalyzing the conversion of androgens to estrogens in humans). Along with Toy et al. [68••], these insights into the different types of P-450 chemistry could lead to new understanding of the enzymes' reactivity.

The authors propose an alternative agostic mechanism to the commonly accepted rebound mechanism (see also [65]). Here, the R-H substrate is suggested to bind to the high-valent oxo-iron intermediate both through the terminal carbon (R) and the hydrogen, weakening the C-H bond, where the oxygen atom insertion occurs. Although the results are only primary, this possibility seems to be consistent with many known results for the P-450s (and the authors expand the relevance of the proposed mechanism to other enzymes). This paper could be a platform to many new experiments in the field.
