By employing paraformaldehyde-fixed, paraffin-embedded sections from both the retinas of both control subjects and patients with primary open-angle glaucoma, these authors describe the existence of two distinct types of astrocytes in the optic nerve head. They show that in primary open-angle glaucoma, type 1B astrocytes exhibit both a greater immunoreactivity to GFAP and N-CAM and an altered morphology. These astrocyte responses are hypothesized to have a pathogenic significance for glaucomatous optic nerve neuropathy.

In order to raise the IOP, chronic ocular hypertension is induced, by occlusion of rat episcleral veins. In vivo experiments were performed in rats by elevating the IOP for 45 minutes to induce ischemia. The retina was assessed to be damaged after 3 days of reperfusion by electroretinography and immunohistochemistry and these effects were counteracted by intraperitoneal injection of betaxolol before ischemia. In vitro experiments showed that betaxolol also prevented changes to rabbit retinas induced by ionotropic glutamate receptor agonists and oxygen/glucose deprivation.

This is a very useful study describing the occurrence of multiple functions for GLAST and GLT-1 glutamate transporters in mice retinas. The authors demonstrate that GLAST is required for normal signal transduction between photoreceptors and bipolar cells and that both GLAST and GLT-1 play a neuroprotective role during ischemia in the retina.
