    • The bHLH protein PTF1-p48 is essential for the formation of the exocrine and the correct spatial organization of the endocrine pancreas
    • This paper reports on the first pancreatic exocrine transcription factor, p48, to be genetically analyzed. The p48 gene encodes the exocrine-specific counterpart of the PTF1 transcription factor complex that previously has been shown to bind to the regulatory region of exocrine-specific genes. p48-null-mutant mice die at the neonatal stage and completely lack exocrine pancreatic tissue. Interestingly, endocrine cells develop in p48-deficient mice but become localized to the spleen. Thus, p48 is the first gene described that selectively impairs exocrine development. This paper also provides genetic evidence that islet cell differentiation, but not organization, is independent of exocrine cell differentiation

    • This paper describes the role that the homeobox gene Hlxb9 plays during pancreas development. Like IPF1/PDX1, Hb9 is transiently expressed in early pancreatic buds, but later in development its expression is restricted to differentiated β-cells. The selective agenesis of the dorsal bud in Hlxb9 mutants represents the first evidence demonstrating a genetic distinction between the development of the dorsal and ventral pancreatic buds. Thus, this study shows that Hlxb9 is required for the initiation of the dorsal pancreatic program and for ensuring proper endocrine cell differentiation in the ventral bud
