Iterative elastic 3D-to-2D alignment method using normal modes for studying structural dynamics of large macromolecular complexes

HEMNMA approach to analyzing continuous conformational changes was introduced in this article. The article also shows performances of the method using several synthetic and experimental data sets. HEMNMA analyzes EM images using normal modes of a starting 3D model obtained by EM or X-ray crystallography. Conformational parameters obtained by image analysis are then used to map images onto a low-dimensional space where conformational changes can be explored (using animations of the starting model or 3D reconstructions along trajectories identified in most densely populated regions in this space).

The method assumes that heterogeneity is not excessive so that each image can be associated with a correct projection direction using standard image-orienting methods that do not take into account conformational heterogeneity.
