Characterization of human immunodeficiency virus type 1 (HIV-1) diversity and tropism in 145 patients with primary HIV-1 infection
-
In this study patients were analyzed for coreceptor tropism and overall variability. For the vast majority of the viruses predicted as CXCR4-using based on the V3 sequence, the coreceptor tropism could not be confirmed in the phenotype analysis. These observations indicate that some of the differences in X4 and R5 prevalence data as reported by different studies are due to differences in the methodology used for tropism determination

Presents the results of retesting of samples from four different cohorts of HIV-1infected patients that have been analyzed for coreceptor tropism with the original Trofile assay Nine to 26% of patients with CCR5-tropic virus by the original Trofile assay had CXCR4-using virus by TF-ES The higher X4 detection rate of the enhanced sensitivity assay was most pronounced in the treatment-experienced population These findings illustrate the importance of test validation 36 Brumme ZL Goodrich J Mayer HB et al. Interleukin-7 induces HIV type 1 R5-to-X4 switch
-
The authors provide in-vitro evidence for the possible involvement of a host factor in the R5 to X4 switch process. They were able to show that IL-7 increases CD4 Tcell surface CXCR4 density, HIV-1 production and the risk of emergence of X4 strain In contrast, HIV-1 production remained low in the absence of IL-7, and no X4 strains were detected. They hypothesize that increased CXCR4 density and higher replication enhance the selection of X4 variants
