By using transgenic mice that constitutively express STAT5b molecules, these investigators demonstrated that homeostatic proliferation of naïve and memory CD8 T cells occurred in mice deficient in both IL-7 and IL-15.

Hancock Homeostatic proliferation is a barrier to transplantation tolerance Nat Med 10 2004 87 92 This paper highlights that an important consequence of homeostatic proliferation is resistance to tolerance induction. Using monoclonal antibodies to induce clinically relevant degrees of sub-total T cell depletion in a mouse model, it was shown that residual non-deleted T cell underwent homeostatic proliferation and acquired phenotypic and functional memory cell characteristics. These cells were able to reject cardiac allografts even in the presence of co-stimulatory blockade.

Strom Favorably tipping the balance between cytopathic and regulatory T cells to create transplantation tolerance Immunity 19 2003 503 514 In this study, the researchers developed a tolerizing protocol to facilitate graft acceptance by combining rapamycin, agonist IL-2 and antagonist IL-15 cytolytic fusion proteins. This protocol selectively deleted alloaggressive lymphocytes while preserving regulatory T cells, and offers future therapeutic development.
