Describes the generation of a transgenic mouse in which the ES cell pluripotency regulator Oct4 is under the control of an inducible promoter, and demonstrates that forced expression of Oct4 in adult mice prevents the terminal differentiation of epithelial progenitor cells, which usually differentiate into gut epithelial cells throughout the life of the adult in a continual cycle of cell sloughing and regeneration. This block to differentiation causes intestinal hyperplasia, and provides a direct link between aberrant expression of an ES-specific gene and abnormal growth.
