    • The intracellular antibody capture technology (IACT): towards a consensus sequence for intracellular antibodies
    • ••], this paper led to the definition of a broadly applicable consensus framework for intrabody library generation. Using the 2-hybrid screening system described in [13] the two groups isolated several panels of intrabodies from diverse phage libraries and analysed the resulting structures. The isolated intrabodies were strikingly similar in their framework structures and allowed the authors to define a minimal sequence that could support intracellular folding.

This finding was slightly counterintuitive, as the general expectation was that the potential for the domains to have large exposed hydrophobic surfaces would make them unsuitable for intracellular expression. The heavy chain of a previously isolated scFv was found to have greater activity than the parent molecule. This led the authors to build a small library of single domains, based on the heavy chain, and isolated new clones from this library which were also superior in behaviour to the originating scFv.
