    • MHC class II-peptide complexes and APC lipid rafts accumulate at the immunological synapse
    • This study makes use of a mAb that recognizes a specific pMHC-II complex to examine the recruitment and distribution of both immunologically relevant and irrelevant MHC-II at the immune synapse. Under conditions of limiting antigen dose, relevant pMHC-II complex, B-cell lipid rafts and even small amounts of irrelevant MHC-II are recruited to the immunological synapse. Co-patching studies using pMHC-II-specific mAb suggest that recruitment is mediated by the association of relevant and irrelevant MHC-II with lipid rafts, providing a potential role for rafts in recruiting un-engaged class II molecules to the T-cell-APC contact site.

    • This study investigates the association of MHC-II, HLA-DM and B7.2 with proteins of the tetraspanin web. They find tetraspanin-associated MHC-II molecules possess a unique, limited peptide repertoire in comparison to the bulk of MHC-II or raft-associated MHC-II. Using a mAb that reportedly associates with tetraspanin-associated MHC-II they show increased surface expression of these complexes during DC maturation, suggesting an important role for tetraspanin web associations in MHC-II-dependent T-cell activation.
