This report is among the first to suggest a mechanism for the activation of a mitochondrial fission or fusion protein, and also contributes to the growing body of literature implicating ER-mitochondrion crosstalk in apoptosis.

    • Using RNAi, the authors illustrate that knockdown of OPA-1 alone leads to dramatic restructuring of the inner mitochondrial membrane, mitochondrial fragmentation, cytochrome c release and apoptosis, suggesting a role for the mitochondrial fission/fusion machinery in normal maintenance of cristae structure and cytochrome c sequestration. This report also implicates apoptosis, resulting from OPA-1 haploinsufficiency, as a pathogenic mechanism in autosomal DOA.

J Biol Chem. 278:2003;7743-7746 Using RNAi, the authors illustrate that knockdown of OPA-1 alone leads to dramatic restructuring of the inner mitochondrial membrane, mitochondrial fragmentation, cytochrome c release and apoptosis, suggesting a role for the mitochondrial fission/fusion machinery in normal maintenance of cristae structure and cytochrome c sequestration. This report also implicates apoptosis, resulting from OPA-1 haploinsufficiency, as a pathogenic mechanism in autosomal DOA.
