Caspase cleavage product of BAP31 induces mitochondrial fission through endoplasmic reticulum calcium signals, enhancing cytochrome c release to the cytosol
-
By manipulating ER-to-mitochondrion calcium signaling, the authors demonstrate that an apoptotic rise in intracellular calcium activates Drp-1 and thus integrates the cell death pathway with the mitochondrial fission machinery. This report is among the first to suggest a mechanism for the activation of a mitochondrial fission or fusion protein, and also contributes to the growing body of literature implicating ER-mitochondrion crosstalk in apoptosis.

HFis1, a novel component of the mammalian mitochondrial fission machinery
-
The authors identify and clone the human orthologue of yeast Fis1p and find it localized at the outer mitochondrial membrane. Evidence is provided that overexpression of hFis1 alone is sufficient to induce mitochondrial fission and apoptosis in cell lines.

Spatial and temporal association of Bax with mitochondrial fission sites, Drp1, and Mfn2 during apoptosis
-
The authors show that Bax colocalizes at the outer mitochondrial membrane with mitochondrial fission/fusion components Drp-1 and Mfn2 during apoptosis, suggesting the involvement of pro-apoptotic Bcl-2 family members in mitochondrial dynamics. By blocking the function of the fission machinery, the authors capture the accumulation of Bax at foci of attempted, but incomplete, mitochondrial scission.

Loss of OPA1 perturbates the mitochondrial inner membrane structure and integrity, leading to cytochrome c release and apoptosis
-
Using RNAi, the authors illustrate that knockdown of OPA-1 alone leads to dramatic restructuring of the inner mitochondrial membrane, mitochondrial fragmentation, cytochrome c release and apoptosis, suggesting a role for the mitochondrial fission/fusion machinery in normal maintenance of cristae structure and cytochrome c sequestration. This report also implicates apoptosis, resulting from OPA-1 haploinsufficiency, as a pathogenic mechanism in autosomal DOA.
