Using a positional candidate approach, this study demonstrates that 5LO protects against aortic lesion formation in two mouse models, even in the presence of elevated cholesterol levels. It was also shown that 5LO is expressed in the cells of atherosclerotic lesions, particularly in macrophage-rich regions.

Arterioscler Thromb Vasc Biol 2003; 23:1072-1076. This study demonstrated that substituting the corresponding amino acid from CON6 mice into the human 5LO protein significantly reduced the activity of the enzyme. These results support the observation that CON6 mice have very low levels of 5LO and LTs.
