    • Rapid recruitment of NMDA receptor transport packets to nascent synapses
    • The authors utilized a time-lapse dual-laser fluorescence microscopy imaging technique to observe behaviors of EGFP- or DsRed-labeled NMDA-Rs, AMPA-Rs and PSD-95 in developing cultured rat cortical neurons. By characterizing mobility and the recruitment patterns of those glutamate receptor-containing transport packets to synaptic sites together with analyzing correlation with the movement of PSD-95, they found rapid recruitment of NMDA-Rs and less dynamic nature of AMPA-Rs. This rapid recruitment of NMDA-Rs explains the dominance of NMDA-Rs in nascent synapses.

    • Applying a multi-electrode stimulation system to hippocampal slices for maximum activation of CA1 synapses in combination with biochemical/immunochemical analyses of surface expression of glutamate receptors, the authors reveal differential surface expression patterns of NMDA-Rs and AMPA-Rs upon LTP induction. The results support a hypothesis that rapid insertion of NMDA-Rs from the intracellular pool to the postsynaptic membrane is a key step in generating LTP.

    • The authors dissociated the interaction sequences of the carboxy-terminal cytoplasmic portion of GluR2 and AP2 or NSF by using mutated GluR2 transgene constructs and specific antagonizing peptides in cultured hippocampal neurons. The results showed that AP2 is crucially important for NMDA-R-mediated internalization of AMPA-Rs at synaptic sites, a potential mechanism of LTD.

They report fast and slow kinetics for reversible scattering/clustering of mGluR5; fast kinetics were confirmed by FRAP of mGluR5-GFP.
