Hence, by analogy to the synapse, SNARE complexes containing non-neuronal VAMP, syntaxin, and SNAP-23 may control Golgi→plasma membrane trafficking in non-neuronal tissues.

The complex is found to be made up of several overlapping subcomplexes with some members (GOS-28 and rbet!) being mutually exclusive and others (syntaxin 5 and rsec22b) being found in common.

A novel syntaxin family member, Ufe1p, was localized to the ER and found to function in the retrieval of the HDEL (single-letter amino acid code) receptor and of proteins containing dibasic retrieval signals from the Golgi back to the ER. Ufe1p was proposed to represent a receptor for retrograde Golgi-derived vesicles. Thus, retrograde ER→Golgi transport appears to involve SNARE mechanisms, and involves a distinct subset of SNARE proteins.

Characterization of a mammalian sequence homolog of Pep12p, termed syntaxin 6, revealed significant sequence homology of syntaxin 6 to SNAP-25 as well. Syntaxin 6 was localized to the Golgi area, consistent with a role in Golgi→endosome transport, as proposed for Pep12p. Syntaxin 6 bound α-SNAP, probably through a predicted coiled-coil domain as for other syntaxin family members.

Nature 1997, 387:199-202. v-SNAREs and t-SNAREs resembling those involved in heterotypic fusion reactions were required for homotypic fusion, demonstrating that similar principles govern vesicle transport and organelle assembly. Although both SNAREs are normally present on vacuolar membranes, vacuoles containing only the v-SNARE (Nyv1p) fuse with vacuoles containing only the t-SNARE (Vam37p). A requirement for SNARE proteins on both fusing membranes was directly demonstrated, verifying a central postulate in the field of membrane trafficking.

A yeast two-hybrid screen using SNAP-25 enabled the authors of this paper to find Hrs-2, a novel 115kDa cytosolic and membrane-associated ATPase. Hrs-2 binds tightly to SNAP-25 in vitro and in vivo, indicating a regulatory role in mechanisms involving SNAREs. Hrs-2 may be involved in calcium-dependent transport phenomena, as its binding to SNAP-25 was calcium- regulated and addition of excess Hrs-2 to permeable neuroendocrine cells inhibited regulated secretion.

Although this would represent a major change in the way we see NSF acting, it remains to be seen whether this homotypic system faithfully represents events in heterotypic fusion.
