Maturation and viability are decreased when degradation of IκB (an inhibitor of NF-κB) and nuclear translocation of NF-κB are prevented. In contrast, an inhibitor of MEK1 (a kinase that activates ERK) does not affect LPS induced maturation but abrogates LPS dependent protection from apoptosis.

    • - mouse DCs. Oval-bumin in immune complexes is presented much more efficiently than soluble protein by either MHC class II or class I molecules. Presentation by MHC class I involves antigen access to the endogenous pathway. These results indicate that DC receptors can serve a dual function in antigen uptake and delivery to processing compartments and in signalling for DC maturation.

Mouse DCs pulsed with HIV ex vivo retain the virus in infectious form and transport it to draining lymph nodes following subcutaneous administration. These observations suggest that DCs may be important for routing infective HIV from peripheral sites to lymph nodes even when the cells are not themselves infected.
