A homologue of Drosophila aurora kinase is oncogenic and amplified in human colorectal cancers

This paper describes the cloning of two human serine/threonine kinase (aurora 1 and aurora 2) related to Drosophila and yeast centrosomal proteins that are required for chromosome segregation. The authors demonstrate that the aurora1 and aurora2 proteins are cell-cycle-regulated and their expression is restricted to mitotic structures. They further demonstrate that aurora2 encodes a transforming protein, and that it maps to an amplicon present in 52% of primary human colorectal cancers.

of outstanding interest. This paper and Taylor 1997 [10] describe the sequence and function of mouse and human Bub1 kinase. Yeast Bub1 is required for a delay in anaphase entry in response to spindle disruption. Similarly, the mammalian counterparts localize to the kinetochore during mitosis, and expression of dominant negative mutants result in failure of cell-cycle arrest following spindle damage. Cahill suggest that Bub1 is involved in a spindle assembly checkpoint, and that a defective Bub1 results in chromosomal instability, a common trait of colorectal tumor cell lines. This study also found 2 of 19 primary colorectal tumors to contain mutant Bub1 alleles, and suggests that the resultant aneuploidy may play a role in tumor progression.

of outstanding interest. This paper describes the cloning and characterization of C. elegans ced-5, a gene required for engulfment of cell corpses during programmed cell death in worms. The protein is similar to human DOCK180, which has been implicated in signaling of cell movement through tyrosine kinases. These findings provide important clues to understanding the molecular events of apoptosis.

An orphan receptor tyrosine kinase family whose members serve as non-integrin collagen receptors

of outstanding interest. This paper and Vogel 1997 [37], demonstrate the direct activation of the DDR1 and DDR2 tyrosine kinase receptors by collagen. Collagen binding results in a delayed autophosphorylation of DDR and subsequent association with SHC, yet no activation of MAP kinase. Vogel [37], also observe that collagen-induced activation of DDR2 results in upregulation of matrix metalloproteinase-1 (MMP1), and suggest this may be a mechanism to promote tumor escape and metastasis.
