This paper examines the effect of ApoE-4 by age and sex in a large (417 cases: 1030 control individuals) case-control study. No differences in the ApoE-4 effect were found in men and women, but the effect of ApoE-4 was estimated to have its strongest effect between the ages of 60 and 79 years.

A sibship test for linkage in the presence of association: The sib transmission/disequilibrium test
-
Spielman RS, Ewens WJ. A sibship test for linkage in the presence of association: the sib transmission/disequilibrium test. Am J Hum Genet 1998; 62:450-458. This recent publication extends the authors' transmission/disequilibrium test for candidate gene analysis to pairs of affected and unaffected siblings. This extension allows a transmission/disequilibrium test-style candidate gene analysis in late-onset diseases (such as Alzheimer's disease), in which parents are unavailable for genotyping.

Complete genomic screen in late-onset familial Alzheimer disease: Evidence for a new locus on chromosome 12
-
Pericak-Vance MA, Bass MP, Yamaoka LH, Gaskell PC, Scott WK, Terwedow HA, et al. Complete genomic screen in late-onset familial Alzheimer disease: evidence for a new locus on chromosome 12. JAMA 1997; 278:1237-1241. This report of a recently completed genomic screen in 54 late-onset Alzheimer's disease families identifies a region on chromosome 12 that is likely to harbor an additional Alzheimer's disease susceptibility gene.

The clinical introduction of genetic testing for Alzheimer disease: An ethical perspective
-
Post SG, Whitehouse PJ, Binstock RH, Bird TD, Eckert SK, Farrer LA, et al. The clinical introduction of genetic testing for Alzheimer disease: an ethical perspective. JAMA 1997; 277:832-836. This most recent consensus statement on genetic testing considers testing for mutations in APP, presenilin-1 and presenilin-2 responsible for autosomal dominant, early-onset Alzheimer's disease, as well as predictive and diagnostic use of APOE genotyping in late-onset.

Ann Neurol 1997; 42:319-325. This neuropathologic study from the Consortium to Establish a Registry for Alzheimer's Disease confirmed the results from other studies [94,96,97], reporting specificity and sensitivity of 83% and a positive predictive value of 97% for the use of the ApoE-4 allele as a test for Alzheimer's disease in patients with a probable diagnosis.
