Torres et al. examined the relationship between Cdc42 and phosphorylation-dependent activation. Their findings indicate that phosphorylation of Tyr256 is unfavorable unless activated Cdc42 is present. They suggest that transient activation of N-WASP by Cdc42 may allow long-term activation via phosphorylation, a form of memory.

This paper and the following one [37] highlight the importance of co-recruitment in determining signaling specificity.

    • •] exemplify new findings indicating that natural scaffold proteins utilize higher order mechanisms of regulation in addition to simple tethering of pathway components. The interactions of the Ras pathway scaffold KSR (kinase suppressor of Ras) are highly regulated. First, signaling activity induces KSR membrane recruitment and interaction with ERK (via displacement of a 14-3-3 protein). Second, signaling activity also relieves repression by the newly identified negative regulator IMP (impedes mitogenic signal propagation), which sequesters KSR in an inactive state. These multiple layers of scaffold regulation are likely to result in a precise, finely tuned Ras pathway response.
