The authors demonstrated that dendritic cells respond to immune complexes by altering their cytokine production such as IL-12p70 to induce a Th2-like cytokine response in T cells. Furthermore, the mechanism by which dendritic cells altered their cytokine production in response to immune complexes was shown to be different from macrophages. Macrophages downregulated the transcription of both the p40 and p35 subunits of IL-12, whereas dendritic cells decreased only p35 expression.

The increased representation of CLIP-MHC class II complexes favored polarization of autologous naive T cells toward the nonpolarized and Th2 phenotype. Thus, the self-peptide CLIP on dendritic cells qualifies as an endogenous regulator in priming of Th cells by antagonizing the polarization toward the Th1 phenotype.

While Th1 cells use multiple suppressive mechanisms, IL-10 and transforming growth factor-β as secreted cytokines, and cytotoxic T-lymphocyte antigen 4 and programmed death 1 as surface molecules, healthy and allergic individuals exhibit all three allergen-specific subsets in different proportions, indicating that a change in the dominant subset may lead to allergy development or recovery. Blocking the suppressor activity of Th1 cells or increasing Th2 cell frequency was found to enhance allergen-specific Th2 cell activation ex vivo.
