-
This investigation established that anthracyclines reduce the number of cardiac progenitor cells in young mice. They go on to demonstrate that the mice then have adverse response to exercise induced cardiac hypertrophy. This study opens up a novel mechanism for cardiotoxicity of anthracyclines, particularly in the heart. It further suggests that strategies that preserve or restore progenitor cell populations may have therapeutic potential for anthracycline-Associated cardiomyopathy

This basic investigation was motivated by effects of multi-tyrosine kinase inhibitors such as sunitinib on cardiac function. Sunitinib inhibits the activity of the PDGF receptor. The investigators show that mice lacking the PDGFR have accelerated heart failure in response to pressure overload. In addition to providing a potential mechanism for the effect of sunitinib on the heart, the study may stimulate reverse translation to examine whether PDGFR signaling can be manipulated to improve outcome in heart failure
