This review article summarizes the central function of activated NF-κ B in response to pro-inflammatory cytokines, excessive mechanical stress, and ECM degradation products in chondrocytes. Activated NF-κ B also regulates the expression of chemokines, adhesion molecules, and several matrix-degrading enzymes. The authors propose that NF-κ B-activating kinases are potential therapeutic OA targets. Therefore, strategies that prevent excessive NF-κ B activation in this context may be used to treat inflammatory joint diseases. Furthermore, exclusive targeting of such strategies at affected cartilage and joints can avoid undesirable systemic effects of NF-κ B inhibition.
