An epigenetic role for maternally inherited piRNAs in transposon silencing

This paper describes how a phenomenon called 'hybrid dysgenesis' is related to piRNA inheritance. The authors found that piRNAs, most probably in a form associated with the PIWI proteins, serve as vectors for epigenetic information from mother, but not from father, to progeny.

A piRNA pathway primed by individual transposons is linked to de novo DNA methylation in mice
-
In this paper, the authors used bioinformatics to reveal differences between MILI-associating and MIWI2-associating piRNAs and found that transposon copies are the origins of primary piRNAs in mice and that MILI prefers to bind primary piRNAs whereas secondary piRNAs are loaded onto MIWI2. The piRNA pathway and de novo methylation was also linked in this study.

Developmentally regulated piRNA clusters implicate MILI in transposon control
-
This study described that MILI-associating piRNAs may regulate retrotransposons through DNA methylation. The amplification loop system for piRNA biogenesis in mouse was also described for the first time in this paper.

Endogenous siRNAs from naturally formed dsRNAs regulate transcripts in mouse oocytes
-
This paper, together with reference [53], was the first to describe the identification of esiRNAs in mouse oocyte. These papers described that esiRNAs regulate not only protein-coding transcripts but also retrotransposons. Some retrotransposons in mice are repressed through both the piRNA and esiRNA pathways.
