    • Reconstitution of circadian oscillation of cyanobacterial KaiC phosphorylation in vitro
    • This landmark paper demonstrates that purified clock proteins KaiA, KaiB and KaiC can mediate circadian phosphorylation of KaiC in vitro. These results show that the core circadian timekeeping mechanism resides in the functions of these three proteins and does not involve rhythmic transcription.

    • Balance between DBT/CKIε kinase and protein phosphatase activities regulate phosphorylation and stability of Drosophila CLOCK protein
    • This study demonstrates that CLK phosphorylation cycles in a circadian manner, but that the overall levels of CLK are constant. In addition, DBT phosphorylates and destabilizes CLK, whereas PP2A dephosphorylates and stabilizes CLK. These results suggest that DBT and PP2A, which were already known to modulate PER phosphorylation, are also important regulators of transcriptional activity.

    • In this paper, mutant BMAL1 and CLOCK proteins are generated that are unable to bind CRY, but activate transcription normally. These mutant proteins are used to show that transcriptional feedback by CRY is required for rhythmic transcription and, thus, circadian oscillator function. The results indicate that rhythmic transcription is essential for circadian oscillator function in mammals.
