    • Production of infectious hepatitis C virus in tissue culture from a cloned viral genome
    • Transfection of in vitro transcripts of the JFH-1 isolate into Huh-7 cells results in secretion of virus that is infectious for naive Huh-7 cells and for a chimpanzee. Infectivity is neutralized by CD81-specific antibodies and, to some extent, by immunoglobulins from chronically infected patients. Immuno-electron microscopy studies reveal spherical particles of about 50 nm. An engineered HCV genome with a stably integrated luciferase reporter gene is described that produces infectious particles.

    • A chimeric virus, composed of the core to NS2 region of J6(CF) and the JFH-1 replicase, was shown to release infectious particles from transfected Huh7.5 cells. • Cardif is an adaptor protein in the RIG-I antiviral pathway and is targeted by hepatitis C virus
    • The authors identify a novel signal transducer that relays the double-stranded RNA-induced signal from retinoic acid inducible gene-I (RIG-I) to the kinase complex responsible for interferon-regulatory factor 3 (IRF-3) phosphorylation. This signal transducer (called 'Cardif' in this report) is proteolytically cleaved by the NS3/4A protease resulting in an inactivation of the RIG-I pathway.

    • Immune evasion by hepatitis C virus NS3/4A protease-mediated cleavage of the toll-like receptor 3 adaptor protein TRIF
    • In a search for the mechanism underlying NS3/4A-mediated inhibition of IRF-3 activation, the authors found that the viral protease causes specific proteolysis of Toll-IL-1 receptor domain-containing adaptor inducing IFN-β (TRIF or TICAM-1), which is an adaptor molecule linking Toll-like-receptor 3 to the kinase complex responsible for activating IRF-3 and nuclear factor (NF)-κB transcription factors.
