By testing the expression of antibody fragments in different protease-deficient strains, this work shows that Prc is the main protease responsible for the recombinant protein degradation and that DegP makes a minor contribution. The production of high levels of the Fab dimer by E. coli fermentation was reported.

Antibodies were fused to the transporter C domain of IgA protease of Neisseria gonorrheae. Hybrid proteins were efficiently translocated regardless of the presence of disulfide bonds. This study was performed in different mutant strains revealing that DsbA and FkpA assist the folding of the N domains in the periplasm.

They demonstrate that disulfide-containing substrates that are expressed in E. coli mutants having an oxidizing cytoplasm can be exported in a Tat-dependent fashion, and suggest the existence of a quality control mechanism intrinsic to the Tat apparatus that determines the competency of folded proteins to be exported. Moreover, this work points to the possible application of the Tat pathway for the production of recombinant proteins in E. coli.

The fusion proteins were shown to exhibit both thiol oxidase and disulfide isomerase activities. In this way a single protein was able to catalyze the formation of native disulfide bonds in heterologous proteins, resulting in native protein yields comparable or better than in bacteria that overexpressed the wild-type DsbC. This work also revealed that the relative orientation of the catalytic domains of disulfide isomerases play a critical role in the rearrangement of disulfide bonds.
