    • IL-15 availability conditions homeostasis of peripheral natural killer T cells
    • -/- mice, suggesting that IL-15 might not be necessary for the development of these cells per se. However, the transfer of carboxyfluorescein diacetate succinimydil diester (CFSE)-labeled Vα14i NKT cells into immunocompromised recipient mice clearly demonstrated that IL-15 was essential to the survival and homeostatic proliferation of Vα14i NKT cells. Furthermore, these studies also suggested that IL-15 availability in vivo might control the size of the Vα14i NKT-cell population. A comparison of Vα14i NKT cell transfers into NK-proficient versus NK-deficient hosts suggests that Vα14i NKT cells and NK cells (and perhaps other IL-15-dependent cell populations) compete for peripheral IL-15.

With the help of transgenesis, CD1d was re-expressed under the control of MHC class II or MHC class I promoters. Neither expression pattern was able to fully restore the Vα14i NKT-cell population. These studies highlight the crucial role played by the cell type, probably the DP cortical thymocytes expressing CD1d, for the positive selection of Vα14i NKT cells.

    • Restoration of NK T cell development in fyn-mutant mice by a TCR reveals a requirement for Fyn during early NK T cell ontogeny
