Development 130, 3891-3902 (2003). The cis-acting sequences that regulate the SHH floor plate enhancer SFPE2 were characterized in this study. The highly conserved binding sites that match the consensus for homeodomain, FOXA and FOXH1 transcription factors were identified. Mutational analysis revealed that the HD and FOXA-binding sites are both required for activation of the SHH floor plate enhancer, but that the FOXH1-binding site is not required. In addition, mutational analysis revealed that a T-box-binding site normally mediates Shh suppression in p4/p5. This study also provides evidence that the floor plate and notochord arise from distinct precursors in the mouse node.
